Use of Random amplified Polymorphic DNAs for Linkage Group Analysis in Interspecific Hybrid F2 Generation of Cucurbita

Abstract
F©ü population of a Cucurbita interspecific hybrid (C. pepo ¡¿ C. moschata) was analyzed to test the use of random amplified polymorphic DNA (RAPD) markers in constructing molecular genetic map. Out of 70 arbitrary 10-mer primers screened, 15 primers showed polymorphisms in parental DNAs and these primers were used to identify the polymorphisms in 40 F©ü individuals. Segregating 47 RAPD markers were analyzed by Mapmaker program and five linkage groups were constructed to contain 28 RAPD marker loci.
